In the process of restoring my truck, I filled and smoothed all the holes in the firewall including the speedometer cable hole. I'm trying to keep the wires running through the engine bay and on the firewall hidden. Looking at the 55-59 service manual, the speedometer cable hole is located high and off-center on the drivers side of the cab. I was thinking of mounting it low and center, near the transmission tunnel.

Will this pose any problems for the speedometer cable? Any recommendations?

As long as the cable does not kink, you could route it any way you wanted. You may need to do some research and get a longer or shorter cable to meet your routing needs.
